Quantitative Analysis of T-Cell Receptor b Variable-Gene Usage in Cutaneous Late-Phase Reactions: Implications for T-Lymphocyte Recruitment in Cutaneous Inflammation

چکیده
To determine if functionally distinct T-lymphocyte (T cell) subsets accumulate in late-phase immunoglobulin E-mediated reactions (LPR), we quantitatively analyzed the immunophenotype and the T-cell receptor b variable-gene (Vb) repertoire of T cells in cutaneous LPR. Peripheral blood and skin biopsies were obtained 6 or 24 h after sensitive subjects were challenged with intradermal injections of grass pollen allergen (Ag) and control (C) solution. The frequency of cells expressing CD3, CD4, CD8, CD45RO, and CD25/mm was determined by immunohistochemistry in nine subjects. Vb usage was assessed by reverse transcription-PCR in five of nine subjects. A significantly greater frequency of CD3 and CD45RO (memory) T cells was detected in Ag sites than in C sites at 24 h after challenge but not at 6 h. The frequency of activated (CD25) and helper (CD4) T cells appeared to be increased in Ag sites as well, though not significantly. Vb6 was the most commonly expressed Vb detected in Ag sites, but it was also detected in accompanying C sites. Vb2 was the most commonly expressed Vb detected in C sites. Sequence analysis in one case revealed Vb expression in a 6-h Ag site to be essentially polyclonal. Our findings suggest that memory T cells with Vb expression similar to that in normal skin accumulate in developing cutaneous LPR. The limited usage of Vb suggests a preferential recruitment or retention of reactive T cells from an endogenous subset of skin-homing T cells with its own skewed Vb repertoire.

متن کاملCC Chemokine Receptor (CCR)4 and the CCR10 Ligand Cutaneous T Cell–attracting Chemokine (CTACK) in Lymphocyte Trafficking to Inflamed Skin
The chemokine thymus and activation-regulated chemokine (TARC; CCL17) is displayed by cutaneous (but not intestinal) venules, and is thought to trigger vascular arrest of circulating skin homing memory T cells, which uniformly express the TARC receptor CC chemokine receptor (CCR)4. Cutaneous T cell-attracting chemokine (CTACK; CCL27), expressed by skin keratinocytes, also attracts cutaneous mem...

The possibility to detect markers of T-cell clonality at the Tcell receptor (TCR) p and y loci in skin biopsy samples has proven to be helpful for the often difficult clinical and immunohistochemical diagnosis of cutaneous T-cell lymphoma (CTCL). However, particularly at the early stage of the neoplastic infiltration, an emerging clonal pattern at Southern may be obscured by the germline TCR co...
متن کاملCutaneous lymphomas and pseudolymphomas: A ten-year study at Emam Reza and Omid hospitals in Mashhad, using immunohistochemical and new classification methods
Background: Cutaneous lymphomas are monoclonal neoplastic proliferations of immune cells most frequently T or B cells that infiltrates skin. Development of new diagnostic methods, particularly those for immunophenotyping, have substantially changed classification of these neoplasms. These reasons prompted us to perform this study. Objective: To re-classify cutaneous lymphomas and pseudoly...
متن کاملImmune surveillance and effector functions of CCR10(+) skin homing T cells.
Skin homing T cells carry memory for cutaneous Ags and play an important sentinel and effector role in host defense against pathogens that enter via the skin. CCR10 is a chemokine receptor that is preferentially expressed among blood leukocytes by a subset of memory CD4 and CD8 T cells that coexpress the skin-homing receptor cutaneous lymphocyte Ag (CLA), but not the gut-homing receptor alpha(4...
